Nice Prison follows the story of Horo Mitsuboshi, a model prisoner at Nobana-shi Prison who has served his sentence for three years. Mitsuboshi is known for his discipline and is an example for the other prisoners. One day, a new guard named Tera Adachi is assigned to the prison.

However, he soon realizes that the guards there are incompetent, and the prisoners do strange things like imitating Spider-Man's upside-down kiss, blowing bubbles, and treating the guards like dogs being trimmed.

As the situation becomes more chaotic, Mitsuboshi appears and everyone suddenly obeys the rules. It turns out that Mitsuboshi intentionally put himself in the prison to fix things after learning the cause of the decline in the quality of the facility. Together with Adachi, he tries to confront the main criminal figure responsible for the problems in the prison.
